Synthesis of phosphonopeptide transition state analogs of HIV-1 protease

Abstract

A focus of the research presented here is on the multi-step synthesis, isolation and characterization of phosphorous containing transition state analogs. Several procedures for the synthesis of the aminophosphonate core structure and side chain dipeptides were explored. These structures were designed to mimic the tetrahedral intermediate formed during the protease catalyzed hydrolysis of polypeptide substrates. As such, they are potential inhibitors of the HIV-1 protease enzyme.
